02. Proper Placement of Sample Containers
Place sample containers directly on metal plates without stacking. Ensure full contact between containers and the metal surface-gaps can hinder heat transfer, compromising the freeze-drying process.
03. Tailor Freezing Parameters to Sample Properties
Different samples (e.g., high-moisture, nutrient-rich, or odor-sensitive materials) require specific minimum temperatures and holding times. For instance, nutrient-dense or fragrant samples benefit from rapid freezing to lock in nutritional value and original aromas.
04. Ensure Complete Freezing Before Vacuum Drying
Samples must be frozen to the set minimum temperature and held for 1–2 hours before initiating vacuum drying. Unfrozen or partially frozen samples risk cracking under vacuum pressure; liquid samples may erupt, leading to freeze-drying failure.
05. Maintain Critical Process Conditions
During operation, keep the condenser temperature below -40℃ and ensure the vacuum pump maintains a stable vacuum level (typically 0.1–0.3 mbar). These parameters are non-negotiable for efficient sublimation and sample preservation.

Sublimation is the core of freeze-drying: the sample chamber plates alternate between cooling and heating to facilitate ice-to-vapor conversion without melting. Familiarize yourself with your equipment's sublimation cycle to monitor progress effectively.
07. Safe Termination of the Freeze-Drying Cycle
Once samples are fully dried, adjust the sample chamber temperature to room temperature. First, close the valve between the vacuum pump and the sample chamber/condenser, then shut down the vacuum pump. Slowly open the vent valve-never fully open it abruptly-to prevent dried samples from being blown away by sudden pressure changes.
08. Pre-Startup Checks
Before each use, verify the refrigeration system is functioning correctly and the vacuum pump oil level is between the two red lines. Routine checks extend equipment lifespan and prevent unexpected downtime.
09. Post-Operation Protocol
After drying, turn off the main power supply. Slowly and fully open the vent valve, then defrost the cold well and drain accumulated water. Proper maintenance ensures consistent performance for future use.
