Spectrophotometry is a method of qualitatively and quantitatively analyzing a substance by measuring the absorbance of light at a specific wavelength or a certain wavelength range. It has the advantages of high sensitivity, easy operation and rapidity, and is the most commonly used experimental method in biochemical experiments. Many substances are measured by spectrophotometry. In a spectrophotometer, when light of different wavelengths is continuously irradiated to a sample solution of a certain concentration, an absorption intensity corresponding to a different wavelength can be obtained.


Important indicators affecting experimental results


Wavelength accuracy and repeatability


Each value of the instrument is measured at a certain wavelength. If the wavelength shown and the actual wavelength deviate by a million miles, then what is the coincidence between the measured value and the true value? The importance of this indicator can be seen. .


Maintenance and maintenance


1. Monochromator. The monochromator is the core part of the instrument and is housed in a sealed box and cannot be disassembled. The selected wavelength should be rotated in a balanced manner, and the unavailability is too strong. In order to prevent the dispersive elements from being exposed to moisture, the monochromator desiccant (silica gel) must be replaced periodically. If the desiccant is found to be discolored, it should be replaced immediately.


2. Absorption pool. The absorption cell must be used correctly and special care should be taken to protect the two optical faces of the absorption cell.


3. Detector. The photoelectric conversion element cannot be exposed for a long time, and strong light irradiation or moisture accumulation should be avoided.


4. When the instrument stops working, the power must be turned off.


5. To avoid dust and contamination of the instrument, cover the dust cover when you stop working.


6. If the instrument does not need to be energized regularly, it should be no less than 20~30min each time to keep the whole machine dry and maintain the performance of electronic components.
